Objective:
The present study was conducted to establish a screening method of waterlogging tolerance in sesame at the germination stage and early seedling stage, and to identify the waterlogging tolerance varieties.
Materials and Methods
Plant materials
Fifty-eight (58) sesame accessions including 56 accessions of Sesamum indicum L. and 2 accessions of Sesamum radiatum Schum. & Thonn. were used in this study, which were collected from Myanmar (Supplemental Table 1). All the accessions were multiplied in a greenhouse at T-PIRC (Tsukuba Plant Innovation Research Center) of University of Tsukuba in 2018 crop season. Seeds were collected and stored at 4°C until the use in the experiment.
Methods of germination assay and stress treatment at seed stage (Laboratory screening)
The experiment was laid out in factorial arrangement with completely randomized design with three replications. Prior to the treatment, seed surface was sterilized with 70 % ethanol for about 5 minutes and rinsed with running water thrice. The seeds were submerged in 1ml tube with distilled water. The waterlogging treatments were established as a function of waterlogging days (3 days, 5 days, 7 days, 9 days and 15 days).) at room temperature. A baseline (control) germination test was done by placing the seeds of each accessions on the moist filter paper (Whatman Filter Papers No. 1) in plastic petri-dishes. Each treatment consisted of 10 seeds and evaluated in 3 replications. After the waterlogging treatment, seeds were gently dried on Kimwipe paper for 2-3 hours to drain excess water and then placed in petri-dishes containing moist paper with distilled water. Petri-dishes were then kept in cultivation chamber maintained at 25±1°C. The seeds were considered germinating when the radical length reached at least 2 mm. To compare waterlogging tolerance of the 58 sesame accessions at the seed stage, the germination rate, radical length, hypocotyl length and seedling fresh weight were evaluated 6 days after completing stress treatment. The survival rate and the following parameters were calculated (Zou et al. 2013).
